Currently i am M1 user. However the line is under my GF name.
Last weekend, I ordered iPhone 6S+ by subscribing Singtel Easy Plan.
I retain my M1 number.

My first ever iphone will be delivered tomorrow (30/09), the Singtel service will be rendered on 1 October. However, my GF and I will go to M1 for transferring the name by this weekend.

Currently I dont have nano sim card. I am using Samsung Note 2. How do I activate the new iphone? Can I just borrow friend's sim card to activate the phone first until I change the M1 line to Singtel?

Please advise.

IIRC, the person delivering the phone will activate the phone for you. You can call SingTel to confirm..

I have previously used a prepaid nano SIM to activate, so any nano SIM should be fine.

This is actually how M1 activate for us in house when we said we do not want to use immediately. They just use their in house nano sim to activate then take it out.
