For a Singapore citizen, do we need to be MACS holder to enroll for this? Or just fill up MDAC within 3 days prior to trip, then do a one time manual immigration then can enroll already?

No need to be macs holder. Just for one trip, u do a one time manual counter verification, THEN your next trip into msia u fill up the mdac and can start using e gates liao. Can't use e gate yet on the same trip u did the verification and enrollment, bear in mind.

I am at JB customs now, I filled up MDAC again today but the e-gates showed "passport verification error". ( I already went through manual immigration last week and registered my fingerprints)

The officer told me because I "registered" for MDAC again today because I filled up the form online, then I need to go through the manual counters again.... To "enroll" again...

Just the manual counters will do. And before that, you have to fill out their online arrival card as well. At this point, not sure if anything has changed with regards to this system.

Propose to lock this thread so that there will not be any replies on this confusing thread title.

Formerly there was a MACS system that allowed entry into Johor via unmanned gates at land CIQ checkpoints if a visitor paid and applied for a pass to do so.
MACS was suspended over 2 years ago.
The current system is coined MACS2.0 (unfortunately, IMO), and requires a one-time enrollment process at the CIQ's manned checkpoint counters after submitting the MDAC 3 days prior to a visit. Subsequent entries via the unmanned gate simply require the visitor to submit the MDAC 3 days prior to each visit. No charges apply.
