Not sure whether u guys can c from the photo.

The below part got those black spots. It is very clear esp in white background regardless of screen brightness.

Is this a defect?

Yes it's defect. That time my screen had a similar issue after they supposedly replaced a new screen but I wasn't satisfied and asked them to change again. But mine was only just one in the middle unlike yours so many. The issue is likely due to the manufacturing of the screen, they placed the LEDs too high up resulting in this halo like effect. Essentially your screen is made of many tiny LEDs at the bottom edge and the light is then diffused using a diffuser to form the entire back light of the LCD screen. Go get it replaced if it's still under apple care warranty.
