Setting up iPhone 6s with Apple Watch

Gurus!

Do I have to unpair Apple Watch to the iPhone 6 first when I start using the Apple Watch with iPhone 6s?

Or setting up the iPhone 6s as a back up is suffice?

You can only pair a watch to one iPhone.
