I suspect my upstairs neighbour has a faulty appliance that is wasting a significant amount of water. I can clearly hear water flowing down the drainage pipe 24 hours a day. Should I politely ask them to check it out to prevent excessive water wastage and high bills?

Initially, I thought it was just hot weather, the air conditioner running, or a water change for their fish tank, but the flow is continuous. I suspect an appliance is faulty that they didn't realise is causing the water to keep draining.

if family and IF theres really running water 24/7 (assuming no accident or mass suicide/murder) AND it doesnt affect you, moi thinks should MYOB

I think you should ask your neighbour for an answer to the question. Sounds of continuous flowing water can cause discomfort, annoyance, or stress for some individuals.

Maybe not be the immediate top one but further up... unless that's the highest floor.
