Osaka city? or you meant Kansai area (Osaka, Kyoto etc) ? if the latter, of course 7days! :o

And on when to visit... it depends on what you want to see, sakura? snow? autumn colours? I will avoid the typhoon season.

Actually, I feel that even 7 days is not enough to explore Kansai Area...since you won't have time for places like Wakayama, Himeji, Okayama...

Therefore, for 7 days, you probably could only do something like this:
Day 1: Singapore to Osaka, stay at Osaka
Day 2: Osaka, stay at Osaka
Day 3: Day trip to Nara, stay at Osaka
Day 4: Day trip to Kobe, stay at Osaka
Day 5: Osaka to Kyoto, stay at Kyoto
Day 6: Kyoto, stay at Kyoto
Day 7: Kyoto to Kansai Airport via Haruka Express train

I guess autumn is always the best time to go ...... Kansai is big ya ? You need more than a week just to glimpse the whole place ....

7 days is about rite to cover the more popular places in Kansai such as Kyoto, Osaka, Nara, Kobe.
itinerary will probably be something like this:
Kyoto - 3 days
Osaka - 2 days
Nara - 1 day
Kobe - 1 day

there are lots of other places to go in Kansai, such as Koyasan, Kinosaki, Kumano, Wakayama, Ise etc, and 7 days is certainly not enough. Kumano region itself took me a whole week and i still feel it is not enough

Nov is a good time to go. do check when is the autumn leave period and coincide ur trip with it. for Kansai, it is usually mid to end Nov if i remember correctly.

If going Osaka only, is 5D4N enough?

Although I don't really understand why would someone choose to only go to Osaka when there are lovely places like Kyoto within 30mins train ride away from Osaka...but if you really only want to go Osaka, then yes. 5D4N is more than enough.

However, this is probably what I would recommend though...
Day 1: Singapore to Kansai Airport, visit Dotonbori area, stay at Osaka
Day 2: Day trip to Kyoto, stay at Osaka
Day 3: Osaka, visit Universal Studios? If not, then go Instant Cup Ramen museum and shopping at Umeda area + Tokyu Hands dept store.
Day 4: Osaka, visit Osaka Aquarium and Osaka Castle
Day 5: Osaka to Kansai Airport.

Well, to me, a "must-see" is actually Kyoto.
But for Osaka itself, I liked Tokyu Hands store (which have branches all over Japan and not just in Osaka), Instant Cup Ramen museum and Dotonbori area.

I also went to the Modern Transportation museum at Bentencho station (nice for train lovers) and Osaka castle (didn't really liked it much, personally prefer Matsumoto castle).

Never did went to Universal Studios nor Aquarium so can't comment on those 2 places.

Will spend 4 days in Kyoto
3 days in OSAKA (pending to do day trip to Kobe & Nara)

Day 1, arrive in Kansai Airport, will take a train to Kyoto
Any recommendation for train and hotel?

Arrive Kyoto:
- Day 1
Shijo Street
Teramachi

- Day 2
Nijo Castle
Nishiki Market

- Day 3
Philosopher's Path
Higashiyama District

Any idea where to squeeze in
