4h if you leave at 1AM. If you leave any time during daylight on a work day, expect to spend 1.5h before even leave LA.
That's was my personal experience, but doubt anything changed last year.
I'm going to be a little less pessimistic. Google Maps says that right now (9:15am on a Tuesday morning), it's a shade under 4 hours from downtown LA to the Bellagio in Las Vegas. In the morning you're going to be going in the opposite direction to most of the commuters, so I don't know, this feels like it might be right.
If you leave LA any time after midday though - yeah, then you're going to be caught in the outbound commuter flow and you're not going to have a good time. If you're going to do Los Angeles to Las Vegas, leave early in the morning - or, even better, stay in Anaheim or Palm Springs the night before.
All of these are non-stop driving times, of course, so make sure you add an hour or two for stopovers.
